To perform comparative assessments of the linear dimensions of the individual components of the object, an eyepiece with a scale or with a grid can be used.
The scale is set in the plane of the field diaphragm of the eyepiece with a magnification of 10x. An eyepiece with a scale is installed in the eyepiece tube. Install the paired eyepiece without a scale in another eyepiece tube. While observing through the eyepiece with a scale, focus on a sharp image of the scale, then use the coarse and fine focus knobs to focus the microscope on a sharp image of the object. Observing the image of the object with the second eye through the eyepiece without a scale, rotate the diopter adjustment ring to achieve a sharp image of the object, while not changing the position of the coarse and fine focus knobs. While observing the image of an object with both eyes, move the eyepiece tubes apart so that the visible fields in the left and right tubes are observed as one, that is, set them in accordance with the eye base. With this setting, the observer will see a sharp image of the object simultaneously with a sharp image of the eyepiece scale, which will make it possible to perform the necessary comparative estimates of the linear dimensions of the structures.
To determine the dimensions of structures in a linear measure (in millimeters or microns), it is necessary to use a special ruler - an object-micrometer (calibration slide). The micrometer object should be placed on the object table instead of the object (on straight microscopes, scale up, on inverted microscopes, scale down). On the scale of the object-micrometer, calibrate the scale of the eyepiece for each objective with which measurements will be performed. To do this, focus the microscope on a sharp image of the scale of the object-micrometer in the plane of the eyepiece and turn the eyepiece in the tube, setting the strokes of both scales in parallel. Determine how many divisions of the object-micrometer fit into the eyepiece scale (with lenses of medium and high magnification) or how many divisions of the eyepiece scale occupies the entire object-micrometer (with lenses of low magnification).
Calculate the division value of the eyepiece scale when working with each lens using the formula:
E=TL/A, where
E - the value of division of the eyepiece scale;
L is the number of divisions of the object-micrometer;
T is the scale division value of the object-micrometer indicated on the object-micrometer;
A is the number of divisions on the eyepiece scale.
